Enzymatic oxalate determination in urine.

چکیده
1THE ACCURATE MEASUREMENT of oxalate concentration in urine presents considerable difficulties. The methods available in the literature are based upon either the direct formation of calcium oxalate precipitate and subsequent oxidimetric titration (1-3) or upon a preliminary ether extraction followed by a titrimetric procedure (4, .5). In a more elaborate method, the extraction with ether is combined with a colonmetric determination (6). Perhaps the most reliable approach devised to date employs radioactive-labeled oxalic acid (7), but this procedure involves special equipment not available in many laboratories. This report describes a rapid method for the determination of oxalic acid in urine utilizing the oxalic decarboxylase from the wood-rot fungus Coilybia velutipes and originally suggested by Dr. H. Shimazono. Oxalic acid is stoichiometrically converted to formic acid and CO2 according to the reaction (8)

Five treatment procedures evaluated for the elimination of ascorbate interference in the enzymatic determination of urinary oxalate.
We evaluated the efficacies of five treatment procedures for eliminating ascorbate interference in the enzymatic determination of urinary oxalate. Aliquots of urine samples, containing different amounts of added ascorbate and oxalate, were individually subjected to ferric chloride, sodium nitrite, sodium periodate, charcoal, or ascorbate oxidase treatment to eliminate ascorbate interference. Ox...
متن کاملAn inexpensive method for sensitive enzymatic determination of oxalate in urine and plasma.
In this simple, sensitive, and rapid enzymatic method for the determination of oxalate in urine or plasma, oxalate oxidase (EC 1.2.3.4) prepared from barley seedlings is used to convert oxalate to carbon dioxide and hydrogen peroxide, which is determined photometrically at 600 nm, with use of horseradish peroxidase, by oxidative coupling of 3-methyl-2-benzothiazoline hydrazine with N,N-dimethyl...
متن کاملSimultaneous Determination of Thiocyanate and Oxalate in Urine using a Carbon Ionic Liquid Electrode Modified with TiO2-Fe Nanoparticles
A carbon ionic liquid electrode (CILE) modified with TiO2-Fe nanoparticles was constructed by incorporating TiO2-Fe nanoparticles into the paste matrix. Under the optimized experimental conditions, using differential pulse voltammetry (DPV), the oxidation of thiocyanate and oxalate were occurred at potentials of 0.740 V and 1.010 V, respectively, at the surface of the modi...
متن کاملMeasurement of urinary oxalate: an enzymatic and an ion chromatographic method compared.
An ion chromatographic and an enzymatic method for determination of oxalic acid in urine were compared on the basis of reliability and practicability. Both methods displayed a within-run imprecision of under 5% and a total imprecision of under 10%. The mean recovery of 0.2 mmol/l sodium oxalate lay close to 100% for both methods. After 80 parallel determinations, both methods produced statistic...
متن کاملDetermination of urinary oxalate with commercially available oxalate oxidase.
For this direct colorimetry of urinary oxalate, commercially available oxalate oxidase (EC 1.2.3.4) is used. The urine is first diluted, to diminish the effect of interfering substances. Analytical recovery of oxalate from urines with five different oxalate concentrations (0.4 to 2.0 mmol/L) ranged from 92 to 109% (mean 99%). The within-day and between-day precision (CV) of the method for a wid...
